Before you start these experiments, you need to check NCBI data base to see if the transcription factor A interacts with the promoter B. If there is no such kind data available, you need to run DNA gel retardation assay or DNase I assay to make sure that the transcription factor A interacts with promoter B specifically. Your plan is an important cell-based assay to confirm that the regulation of promoter B transcriptional activities by transcription factor A in tissue cultures.
