抛砖引玉。我先来讲一些基础的,大家补充。
先说一下recA重组酶和recA同源重组系统。理解了recA重组酶的作用,就好理解这个kit的原理和操作方法了。那么就晓得能不 能改进和DIY了。嘿嘿,按mylab战友的说法,可以单独购买试剂节约银子了。 recA介导的同源重组是原核生物DNA修复功能的主要执行者。recA蛋白是一个39ku的单链蛋白,其主要作用是促进DNA同源片段的 配对以及DNA分子间的单链交换,并形成稳定的重组产物。但对于加与反应的底物DNA分子具备在个条件:1,DNA分子间或分子内 存在较高的同源序列,2,至少一种DNA底物是单链,3,至少一条DNA链具有自由末端。 在中性pH条件下,recA能大量结合于单链DNA,每个单体与单链DNA上的3-5个碱基结合,而且这种结合作用呈现高度的协同效应 。此外,recA还具有依赖pH和ATP的双链DNA结合活性。 The RecA protein used in the RecActive Kit is a multi-functional E. coli DNA-binding protein that plays integral roles in both homologous recombination and post-replicative DNA repair. RecA catalyzes the pairing of single- stranded DNA (ssDNA) with complementary regions of double-stranded DNA (dsDNA). RecA monomers first polymerize to form a helical filament around ssDNA. Duplex DNA then binds to the polymer. Bound dsDNA partially unwinds to facilitate base pairing between ssDNA and duplexed DNA. Once ssDNA has hybridized to a region of dsDNA, the duplexed DNA further unwinds to allow for branch migration. RecA has a binding site for ATP, the hydrolysis of which is required for release of the DNA strands from RecA filaments. 如果没有recA蛋白,探针杂交可以执行,但就达不到“The use of homologous recombination enzymes and kinetically trapped multi-stranded DNA hybrids enablesDNA to be targeted in its natural, non-denatured, double-stranded state at virtually any DNA sequence”的效果了。变性转移到纤维素膜上的的全长cDNA怎么还能进行后续的转化、克隆实验 呢?嘿嘿,看来,这个东东还是要保留的。 有兴趣的战友再看看这个protocol吧。 recActive.rar(277.48k) |
