What kind of DNA is it, plasmid or genomic sample? How many microgram DNA is used in the digestion? You may need to run agarose gel (1%) to examine the digestion.

Try to cut 5 ug of genomic DNA in 1 x CutSmart buffer and in a fina volumes of 50 ul at 37 C overnight.

Run 20 ul of digested DNA on a 1 % agarose gel in 1 X TAE at 60 V for 2 hours ( for over 10 cm long gel). Take picture under UV light box.

NEB的这个酶理论上1ul能够在15min内切开1ug质粒的，所以按你的酶切体系应该能够完全切开
