脂质体转染原理
　　脂质体是磷脂分散在水中时形成的脂质双分子层，又称为人工生物膜。最初，人们只是运用脂质体模拟生物膜，研究膜的构造及功能，从而发现了膜的融合及内吞作用，因而可用作外源物质进入细胞的载体。
　　阳离子脂质体表面带正电荷，能与核酸的磷酸根通过静电作用将DNA分子包裹人内，形成DNA一脂复合体，也能被表面带负电荷的细胞膜吸附，再通过膜的融合或细胞的内吞作用，偶尔也通过直接渗透作用，将
　　DNA传递进入细胞，形成包涵体或进入溶酶体 其中一小部分DNA能从包涵体内释放，并进入细胞质中，再进一步进入核内转录、表达。
　

　已有众多的文献报道，脂质体本身会参与细胞生理活动,引起基因表达的上调或下调。如参与PKC（蛋白激酶C）通路调节(Biochemistry.1992 Sep 22;31(37): 9025-30)；如抑制ATP酶的活性(Biochim BiophysActa.2008 Apr;1777(4):362-8)；如与线粒体膜发生作用(J Biol Chem. 1989 Jan25;264(3):1508-15)；转染siRNA造成脱靶效应等等。细胞毒性的大小往往意味着对细胞生理活动影响的大小，脂质体这些作用是造成细胞毒性的根本原因。这会对相关研究数据产生严重的干扰，甚至影响研究的结论。这已引起了许多研究者的注意。
　虽然人们早已发现脂质体对细胞的毒 性，对研究的影响，但由于一直没有更好的方法尤其是更高效的转染方法代替脂质体，所以脂质体转染试剂一度应用非常广泛。同时，人们也一直在寻找更有效、毒性小同时对研究影响也小的转染试剂。由于脂质类转染试剂对细胞的毒性是由其脂质特性决定的，目前众多的研究者和生物公司将新一代转染试剂的开发聚焦在非脂质体的聚合物上，并已有一些优秀的试剂产品上市。
