博莱霉素

Zeocin 可选择Sh ble基因表达的细胞。与现在使用的其他动物细胞标记物无交叉抗性。因此这种抗生素可用来分离对其他筛选剂（如：庆大霉素，潮霉素）有抗性的克隆。Zeocin 是一种属于争光霉素家族的糖蛋白抗生素，在体内能作用于大多数细菌（包括E。coli）、真菌（如：酵母菌）、植物细胞、动物细胞、细胞培养级。　　
使用:
　　●液体Zeocin 可用于细胞培养。
　　●用于筛选转化株的 Zeocin  浓度根据pH值和盐浓度改变。
　　●pH越高，盐浓度越低 Zeocin  活性越大。
　　●普通大肠杆菌菌株筛选可在25＆micro；g／ml Zeocin 浓度的低盐LB琼脂培养基中进行，pH调至7.5。
　　●动物细胞筛选：根据不同细胞系从100到750＆micro；g／ml。
　　●植物细胞筛选：根据不同的细胞种类和培养基从50到250＆micro；g／ml。
　　●酵母筛选：取决于种属和培养基，S。cerevisiae Sh ble转化株在50＆micro；g／ml浓度的YPD培养基中筛选，pH7.0.304015.

Description
Zeocin™ Reagent is a member of the bleomycin antibiotic family. Resistance is conferred by the Sh ble gene which encodes a 13,665 dalton protein. In cells expressing the protein, Zeocin™ is prevented from binding and cleaving the cellular DNA. Zeocin™ is effective on most aerobic cells and can be used for selection in  mammalian     and insect cell lines, yeast, and bacteria. The amount of Zeocin ™ required for selection will vary between 50-2000 ug/ml (typically 300 ug/ml) depending on the cell type.
