TNFRSF14, also known as Herpesvirus entry mediator (HVEM), TR2 (TNF receptor like molecule) and ATAR (another TRAF associated receptor), is a type I membrane protein belonging to the TNF/NGF receptor superfamily. The extracellular domain of HVEM contains several cysteine rich repeats characteristic of TNF receptor superfamily members. The short cytoplasmic region lacks a death domain present in some TNF receptor family members, but contains a TRAF (TNF receptor associated factor) interaction domain. The extracellular domain of HVEM has been shown to interact directly with the herpes simplex virus(HSV) envelope glycoprotein D which plays an important role in HSV pathogenesis. Two TNF superfamily ligands, including the secreted TNFβ (lymphotoxin α) and the membrane protein LIGHT competes with HSV glycoprotein D for HVEM.The cytoplasmic region of HVEM was found to bind to several TRAF family members, which may mediate the signal transduction pathways that activate the immune response

·Quality Control
Biological activity:The ED50 of rHuTNFRSF14 is typically 2.5-10μg/mL in the presence of 1 ng/mL of rhTNFß, as measured by its ability to inhibit TNFß mediated cytotoxicity using L929 mouse fibrosarcoma cells.
Purity: Greater than 90.0% as determined by: (a) Analysis by RP-HPLC. (b) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel. Amino-Acid Sequence: Predicted N terminal amino acid is Leu39.
Endotoxin:Less than 0.1ng/μg (1EU/μg) of rHuTNFRSF14.
·Formulation Sterile and Lyophilized from a 0.2 μm filtered solution in PBS.
·ReconstitutionIt is recommended to reconstitute the lyophilized rHuTNFRSF14 in sterile water not less than 100μg/ml, which can then be further diluted to other aqueous solutions.
·Storage Lyophilized rHuTNFRSF14 although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution rHuTNFRSF14 should be stored at 4°C between 2-7 days and aliquots of reconstituted samples containing 0.1% BSA are stable up to 3 months below -18°C for future use.
